Activated Dimethicone or Simethicone, USP is an antifoaming agent that is used to reduce bloating from excessive gas. Activated Dimethicone IP (Simethicone IP) an active ingredient in many antacids, anti - flatulent preparations that causes small air bubbles in the intestines to coalesce and pass out easily. Can be easily incorporated into various Antacid - Anti Flatulent pharmaceutical formulations such as tablets & suspensions. It also finds its application in distilleries & food processing industries.

Specifications of Simeticone BP Ph Eur Grade:
Action and use: Silicon dioxide analogue; defoaming agent.

DEFINITION
Simeticone is prepared by incorporation of 4 per cent to 7 per cent silica into poly (dimethylsiloxane) with a degree of polymerization between 20 and 400.
Content: 90.5 per cent to 99.0 per cent of poly(dimethylsiloxane).

PRODUCTION
Poly(dimethylsiloxane) is obtained by hydrolysis and polycondensation of dichlorodimethylsilane and chlorotrimethylsilane and the silica is modified at the surface by incorporation of methylsilyl groups.

CHARACTERS
Appearance: Viscous, greyish-white, opalescent liquid.
Solubility: Practically insoluble in water, very slightly soluble or practically insoluble in anhydrous ethanol, practically insoluble in methanol, partly miscible with ethyl acetate, with methylene chloride, with methyl ethyl ketone and with toluene.

IDENTIFICATION
A. Infrared absorption spectrophotometry.
B. Heat 0.5 g in a test-tube over a small flame until white fumes begin to appear. Invert the tube over a 2nd tube containing 1 ml of a 1 g/l solution of chromotropic acid, sodium salt in sulphuric acid so that the fumes reach the solution. Shake the 2nd tube for about 10 seconds and heat on a water-bath for 5 minutes. The solution is violet.
C. The residue obtained in the test for silica under Assay, gives the reaction of silicates.

TESTS
Acidity: To 2.0 g add 25 ml of a mixture of equal volumes of anhydrous ethanol and ether previously neutralized to 0.2 ml of bromothymol blue solution and shake. Not more than 3.0 ml of 0.01 M sodium hydroxide is required to change the color of the solution to blue.
Defoaming activity:
Foaming solution: Dissolve 5.0 g of docusate sodium in 1 liter of water, warm to 50C if necessary.
Defoaming solution: To 50 ml of methyl ethyl ketone add 0.250 g of the substance to be examined, warm to not more than 50C with shaking. Into a 250 ml cylindrical tube about 5 cm in diameter introduce 100 ml of foaming solution and 1 ml of defoaming solution. Close tightly and fix the tube on a suitable oscillating shaker that complies with the following conditions:
— 250-300 oscillations per minute;
— angle of oscillation of about 10;
— oscillation radius of about 10 cm.
Shake for 10 seconds and record the time between the end of the shaking and the instant the 1st portion of foam-free liquid surface appears.
This duration is no longer than 15 seconds.
Mineral oils: Place 2.0 g in a test-tube and examine in ultraviolet light at 365 nm. The fluorescence is not more intense than that of a solution containing 0.1 ppm of quinine sulphate in 0.005 M sulphuric acid examined in the same conditions.
Phenylated compounds: To pass the test.
Heavy metals: Maximum 5 ppm.
Volatile matter: Maximum 1.0 per cent, determined on 1.00 g by heating in an oven at 150C for 2 h. Carry out the test using a dish 60 mm in diameter and 10 mm deep.


Specifications of Simethicone USP NF Grade:
-(Trimethylsilyl)- -methylpoly[oxy(dimethylsilylene)], mixture with silicon dioxide ---8050-81-5.

Simethicone is a mixture of fully methylated linear siloxane polymers containing repeating units of the formula [–(CH3)2SiO–]n, stabilized with trimethylsiloxy end-blocking units of the formula [(CH3)3 SiO–], and silicon dioxide. It contains not less than 90.5 percent and not more than 99.0 percent of polydimethylsiloxane ([–(CH3)2SiO–]n), and not less than 4.0 percent and not more than 7.0 percent of silicon dioxide.

Packaging and storage: Preserve in tight containers.
Identification: Infrared Absorption.
Defoaming activity:
Foaming solution: Dissolve 1 g of octoxynol 9 in 100 mL of water.
Test preparation: Transfer 200 mg of Simethicone to a 60-mL bottle, add 50 mL of tertiary butyl alcohol, cap the bottle, and shake vigorously. [NOTE—Warm slightly, if necessary, to dissolve.]
Procedure: [NOTE—For each test, employ a clean, unused, 250-mL glass jar.] Add, dropwise, 500 microL of the Test preparation to a clean, unused, cylindrical 250-mL glass jar, fitted with a 50-mm cap, containing 100 mL of the Foaming solution. Cap the jar and clamp it in an upright position on a wrist-action shaker. Employing a radius of 13.3 ± 0.4 cm (measured from center of shaft to center of bottle), shake for 10 seconds through an arc of 10 degrees at a frequency of 300 ± 30 strokes per minute. Record the time required for the foam to collapse. The time, in seconds, for foam collapse is determined at the instant the first portion of foam-free liquid surface appears, measured from the end of the shaking period. The defoaming activity time does not exceed 15 seconds.
Loss on heating: Heat about 15 g, accurately weighed, in an open, tared vessel having a diameter of 5.5 ± 0.5 cm and a wall height of 2.5 ± 1.0 cm at 200 in a circulating air oven for 4 hours, and allow to come to room temperature in a desiccator before weighing: it loses not more than 18.0% of its weight.
Heavy metals: To pass the test (5 microg per g).
Content of silicon dioxide: To pass the test.


Simethicone Emulsion USP NF Grade:

Simethicone Emulsion is a water-dispersible form of Simethicone composed of Simethicone, suitable emulsifiers, preservatives, and water. It may contain suitable viscosity-increasing agents. It contains an amount of polydimethylsiloxane ([–(CH3)2SiO–]n) that is not less than 85.0 percent and not more than 110.0 percent of the labeled amount of simethicone.

Packaging and storage: Preserve in tight containers.
Identification: The IR absorption spectrum.
Microbial limits: Its total aerobic microbial count does not exceed 100 cfu per g.
