Wool Fat or Lanolin

Wool Fat or Lanolin or Wool Yolk or Wool Wax or Sheep Grease is a wax secreted by wool-bearing animals. Lanolin and its derivatives are used extensively in both the personal care and health care sectors such as topical liniments. Lanolin is used commercially in many industrial products ranging from rustproof coatings to lubricants.

General Properties and Specifications of Wool Fat or Lanolin or Wool Yolk or Wool Wax or Sheep Grease:
Appearance: Translucent, pale yellow, soft tenacious solid.
Odor: Slight characteristic odor or practically odorless.
Relative Density: about 0.935
Melting Point: 38C to 42C literature.
Flash Point: 113C.
Solubility: Practically insoluble in water. Sparingly soluble in cold alcohol and more in hot alcohol. Freely soluble in chloroform, ether.


Lanolin USP Grade Specifications:

DEFINITION
Lanolin is the purified, wax-like substance from the wool of sheep, Ovis aries L. (Fam. Bovidae), that has been cleaned, decolorized, and deodorized. It contains NMT 0.25% of water. It may contain NMT 0.02% of a suitable antioxidant.

IMPURITIES
Residue on Ignition: NMT 0.1%
Chloride and Sulfate, Chloride: To pass the test. --- 0.035%
Foreign Substances: To pass the test.
Melting Range or Temperature:
Analysis: Determine on a sample previously cooled to 8C to 10C.
Acceptance criteria: 38C to 44C.
Fats and Fixed Oils, Acid Value (Free Fatty Acids):
Sample: 10.0 g. Acceptance criteria: The free acids obtained from the Sample require NMT 2.0 mL of 0.10 N sodium hydroxide for neutralization.
Fats and Fixed Oils, Iodine Value:
Sample: 780–820 mg.
Acceptance criteria: 18–36
Alkalinity:
Sample: 2.0 g.
Analysis: Dissolve the Sample in 10 mL of ether and add 2 drops of phenolphthalein TS.
Acceptance criteria: The liquid is not colored red.
Water-Soluble Acids and Alkalies:
Sample: 10.0 g.
Analysis: Warm the Sample with 50 mL of water on a steam bath, constantly stirring the mixture until the Lanolin is melted.
Acceptance criteria: The fat separates completely on cooling, leaving the water layer nearly clear and neutral to litmus. Retain the water layer for the test for Water-Soluble Oxidizable Substances and Ammonia.
Water-Soluble Oxidizable Substances:
Sample solution: 10 mL of the solution from Water-Soluble Acids and Alkalies.
Analysis: Add the Sample solution to 50 µL of 0.10 N potassium permanganate.
Acceptance criteria: The resulting solution does not completely decolorize within 10 min.
Ammonia:
Sample solution: 10 mL of the solution from Water-Soluble Acids and Alkalies.
Analysis: Add 1 mL of 1 N sodium hydroxide to the Sample solution, and boil.
Acceptance criteria: The vapors do not turn red litmus to blue.
Water Determination:
Acceptance criteria:  NMT 0.25%
Petrolatum:
Sample: 3 g.
Analysis: Heat the Sample on a steam bath, with frequent stirring, until its weight loss is NLT its water content. Boil 40 mL of dehydrated alcohol with 500 mg of the dried lanolin so obtained.
Acceptance criteria: The solution is clear or NMT opalescent.
Packaging and Storage: Preserve in well-closed containers, preferably at controlled room temperature.


Wool Fat BP Ph Eur Grade Specifications:
Anhydrous Lanolin

DEFINITION
Purified, anhydrous, waxy substance obtained from the wool of sheep (Ovis aries). A suitable antioxidant may be added.

CHARACTERS
Appearance: Yellow, unctuous substance. When melted, it is a clear or almost clear, yellow liquid. A solution in light petroleum is opalescent.
Solubility: Practically insoluble in water, slightly soluble in boiling anhydrous ethanol.
Characteristic odour.

IDENTIFICATION
A
. In a test-tube, dissolve 0.5 g in 5 mL of methylene chloride and add 1 mL of acetic anhydride and 0.1 mL of sulfuric acid. A green colour develops.
B. Dissolve 50 mg in 5 mL of methylene chloride, add 5 mL of sulfuric acid and shake. A red colour develops and an intense green fluorescence appears in the lower layer when examined in daylight, with illumination from behind the observer.

TESTS
Water-soluble acid or alkaline substances: To pass the test.
Water-absorption capacity: To pass the test.
Acid value: Maximum 1.0, determined on 5.0 g dissolved in 25 mL of the prescribed mixture of solvents.
Peroxide value: Maximum 20.
Saponification value: 90 to 105, determined on 2.00 g while heating under reflux for 4 h.
Water-soluble oxidisable substances: To pass the test.
Paraffins: Maximum 1.0 per cent.
Pesticide residues: To pass the test. Maximum 0.05 ppm for each organochlorine pesticide, 0.5 ppm for each other pesticide and 1 ppm for the sum of all the pesticides.
Isolation of the pesticide residues: To pass the test.
Identification and quantification of the pesticide residues: To pass the test.
Chlorides: Maximum 150 ppm.
Loss on drying: Maximum 0.5 per cent, determined on 1.000 g by drying in an oven at 105C for 1 h.
Sulfated ash: Maximum 0.15 per cent.
Storage: At a temperature not exceeding 25C.

Lanolin, Anhydrous, occurs as a purified, yellow-white, semisolid, fatlike substance. It is extracted from the wool of sheep. It is insoluble in water, but mixes with about twice its weight of water without separation. It is soluble in chloroform and in ether.
Function: Masticatory substance in chewing gum base.

REQUIREMENTS
Acid Value: Not more than 1.12.
Iodine Value: Between 18 and 36.
Lead: Not more than 3 mg/kg.

Inhalation: Remove to fresh air. If not breathing, give artificial respiration. If breathing is difficult, give oxygen. Get medical attention.
