DEFINITION
1,3,7-Trimethyl-3,7-dihydro-1H-purine-2,6-dione.
Content: 98.5 per cent to 101.5 per cent (dried substance).

CHARACTERS
Appearance: White or almost white, crystalline powder or silky, white or almost white, crystals.
Solubility: Sparingly soluble in water, freely soluble in boiling water, slightly soluble in ethanol (96 per cent). It dissolves in concentrated solutions of alkali benzoates or salicylates.
It sublimes readily.

A. Melting point 234C to 239C.
B. Infrared absorption spectrophotometry.
C. To 2 mL of a saturated solution add 0.05 mL of iodinated potassium iodide solution. The solution remains clear. Add 0.1 mL of dilute hydrochloric acid; a brown precipitate is formed. Neutralise with dilute sodium hydroxide solution, the precipitate dissolves.
D. In a ground-glass-stoppered tube, dissolve about 10 mg in 0.25 mL of a mixture of 0.5 mL of acetylacetone and 5 mL of dilute sodium hydroxide solution. Heat in a water-bath at 80C for 7 min. Cool and add 0.5 mL of dimethylaminobenzaldehyde solution. Heat again in a water-bath at 80C for 7 min. Allow to cool and add 10 mL of water; an intense blue colour develops.
E. Loss on drying. Maximum 0.5 per cent, determined on 1.000 g by drying in an oven at 105C for 1 h.
F. It gives the reaction of xanthines.

TESTS
Solution S: Dissolve 0.5 g with heating in 30 mL of carbon dioxide-free water prepared from distilled water, cool and dilute to 50 mL with the same solvent.
Appearance of solution: Solution S is clear and colourless.
Acidity: To 10 mL of solution S add 0.05 mL of bromothymol blue solution; the solution is green or yellow. No more than 0.2 mL of 0.01 M sodium hydroxide is required to change the colour of the indicator to blue.
Related substances: To pass the test by chromatography.
Sulfates: Maximum 500 ppm, determined on 15 mL of solution S.
Loss on drying: Maximum 0.5 per cent, determined on 1.000 g by drying in an oven at 105C for 1 h.
Sulfated ash: Maximum 0.1 per cent, determined on 1.0 g.

DESCRIPTION
Caffeine occurs as a white powder or as white, glistening needles, usually matted together. It may be compacted or compressed into free-flowing granules or pellets. It is odorless and has a bitter taste. Caffeine is anhydrous or contains one molecule of water of hydration. Its solutions are neutral to litmus. The hydrate is efflorescent in air, and 1 g is soluble in about 50 mL of water, in 75 mL of alcohol, in about 6 mL of chloroform, and in 600 mL of ether.
Function: Flavoring agent.

REQUIREMENTS
Labeling: Indicate whether it is anhydrous or hydrous.
Identification:
A. Dissolve about 5 mg of sample in 1 mL of hydrochloric acid contained in a porcelain dish, add 50 mg of potassium chlorate, and evaporate on a steam bath to dryness. Invert the dish over a vessel containing a few drops of 6 N ammonium hydroxide. The residue acquires a purple color, which disappears on the addition of a solution of a fixed alkali.
B. The infrared absorption spectrum of a mineral oil dispersion of the sample, previously dried at 80° for 4 h, exhibits relative maxima at the same wavelengths as those of a similar preparation of USP Caffeine Reference Standard.
Assay: Not less than 98.5% and not more than 101.0% of C8H10N4O2, calculated on the anhydrous basis.
Lead: Not more than 1 mg/kg.
Melting Range Anhydrous: Between 235° and 237.5°.
Other Alkaloids: Passes test.
Readily Carbonizable Substances: Passes test.
Residue on Ignition: Not more than 0.1%.
Water Anhydrous: Not more than 0.5%; Hydrous: Not more than 8.5%.
