Action and use: Antacid; vaccine adjuvant.

DEFINITION
Hydrated AlPO4 in gel form.
Content: 19.0 per cent to 21.0 per cent of AlPO4.

CHARACTERS
Appearance: Gel.
Solubility: Practically insoluble in water, in ethanol (96 per cent) and in methylene chloride. It dissolves in dilute solutions of mineral acids.

IDENTIFICATION
A. Solution S (see Tests) gives reaction (b) of phosphates.
B. Solution S gives the reaction of aluminium.
C. It complies with the assay.

TESTS
Solution S: Dissolve 2g in dilute hydrochloric acid and dilute to 100 mL with the same acid.
pH: 6.0 to 8.0.
Peroxides: Maximum 150 ppm, expressed as hydrogen peroxide.
Chlorides: Maximum 500 ppm.
Soluble phosphates: Maximum 0.5 per cent, expressed as PO4.
Sulfates: Maximum 0.2 per cent.
Soluble aluminium: Maximum 50 ppm.
Arsenic: Maximum 1 ppm, determined on 1.0 g.
Acid neutralising capacity: Add 2g to 30 mL of 0.1 M hydrochloric acid heated to 37C and maintain at 37C while shaking. Determine the pH after 15 min. The pH of the mixture is 2.0 to 2.5.
Residue on ignition: 19.0 per cent to 23.0 per cent.
Microbial contamination:
TAMC: acceptance criterion 1000 CFU/g.
TYMC: acceptance criterion 100 ;CFU/g.
Absence of bile-tolerant gram-negative bacteria.

Aluminum Phosphate Gel is a water suspension containing not less than 4.0 percent and not more than 5.0 percent (w/w) of aluminum phosphate (AlPO4). It may contain sodium benzoate, benzoic acid, or other suitable agent in an amount not exceeding 0.5 percent, as a preservative.

Identification:
A: A solution of it in hydrochloric acid meets the requirements of the tests for Aluminum.
B: A solution of it in 2 N nitric acid meets the requirements of the tests for Phosphate.

pH: between 6.0 and 7.2.
Soluble phosphate: To pass the test. The soluble phosphate, calculated as PO4 does not exceed 0.30%.
Sulfate: --- not more than 0.05% is found.
Arsenic: Prepare the Test Preparation by dissolving 5.0 g of Gel in the smallest necessary volume of 3 N hydrochloric acid. The limit is 0.6 ppm.
Chloride: --- Not more than 0.16% of chloride is found.
