Colchicine is an alkaloid contained in various species of Colchicum and in other genera. It contains not less than 94.0 percent and not more than 101.0 percent of C22H25NO6, calculated on the anhydrous, solvent-free basis.
[Caution— Colchicine is extremely poisonous.]
Packaging and storage— Preserve in tight, light-resistant containers.

Identification: Infrared Absorption.
Specific rotation: between −240° and −250°, calculated on the anhydrous, solvent-free basis.
Test solution: 10 mg per mL, in alcohol.
Water Determination: not more than 2.0%.
Limit of colchiceine: To 5 mL of a solution (1 in 100) add 2 drops of ferric chloride TS: no definite green color is produced.
Limit of ethyl acetate: To pass the test --- not more than 8.0% is found.
Residual solvents: meets the requirements, except that the limit of chloroform is 100 ppm.

Appearance: Yellowish-white, amorphous or crystalline powder.
Solubility: Very soluble in water, rapidly recrystallising from concentrated solutions as the sesquihydrate, freely soluble in ethanol (96 per cent), practically insoluble in cyclohexane.

A. Ultraviolet and visible absorption spectrophotometry.
B. Infrared absorption spectrophotometry.
C. To 0.5 mL of solution S (see Tests) add 0.5 mL of dilute hydrochloric acid and 0.15 mL of ferric chloride solution. The solution is yellow and becomes dark green on boiling for 30 s. Cool, add 2 mL of methylene chloride and shake. The organic layer is greenish yellow.
D. Dissolve about 30 mg in 1 mL of ethanol (96 per cent) and add 0.15 mL of ferric chloride solution. A brownish red colour develops.

Solution S: Dissolve 0.10 g in water and dilute to 20 mL with the same solvent.
Appearance of solution: Solution S is clear and not more intensely coloured than reference solution.
Acidity or alkalinity: To 10 mL of solution S add 0.1 mL of bromothymol blue solution. Either the solution does not change colour or it becomes green. Not more than 0.1 mL of 0.01 M sodium hydroxide is required to change the colour of the indicator to blue.
Specific optical rotation: -235 to -250 (anhydrous substance).
Dissolve 50.0 mg in ethanol (96 per cent) and dilute to 10.0 mL with the same solvent.
Related substances: To pass the test.
